If you want to just get a fastpass, this can be done at the gate. We upgraded to the VIP ticket, and emailed Universal about our wishes and they phoned us back that evening. Their customer service is very good!
You can purchase Express Passes online, through the app, at the parks from the Universal Studios Store, or from Guest Services. They do have a finite amount of Express Passes available each day, and on busier days they will sell-out very quickly, so buy them as soon as you know you want/need them.
