This will be a comprehensive, step-by-step guide to Undertale 's Genocide / No Mercy route. There are lots of guides to the No Mercy route you can find online, but most lack finer details and comprehensive explanations (even the ones that claim to cover such topics).
1. Dark Ruins. Try to kill Decibat, as many Rorrims and Crispy Scrolls as possible for more EXP, don't exhaust the kill counter early. Reach the corn maze, make sure to retrieve the Pebble Ammo (+3 AT) by going around the maze and reaching the top left corner.
An easy method is (CENTER, UP, CENTER, UP). Now the fight begins. The 2nd attack is simply pressing the UP key in rhythm, just be sure to watch the bone slots and don't press the key too long or short. I'd say the hardest attack to get down is the 3rd Attack, the one with the long blue bones and short white bones.
I don't want the whole Offtale Genocide Run to be fighting Secretaries and then just slaughtering New Home. I mean, come on. There's gotta be some interaction with the canon Undertale universe before Asgore.

The Genocide Route occurs when the protagonist kills every enemy (including mini, mid and main bosses) in every region: the Ruins, Snowdin Forest, Waterfall, and Hotland (the CORE is a sub-region of Hotland). Starting in the Ruins, the protagonist repeatedly triggers encounters to kill all monsters.
