10 kwi 2023 · Objectives: Explain the mechanism of action of tamoxifen. Identify the approved indications of tamoxifen, and cite some of the non-approved indications as well. Review the adverse event profile of tamoxifen therapy.
17 lis 2021 · This study demonstrated the feasibility of therapeutic drug monitoring (TDM)-guided tamoxifen dosing. TDM offers the possibility of safely halving the number of patients with subtherapeutic endoxifen levels without introducing additional toxicity.
Tamoxifen has been tested for carcinogenicity by oral and subcutaneous administration to adult and infant mice and rats, and by transplacental exposure to mice.
15 maj 2018 · Purpose: Levels of endoxifen, the most active metabolite of tamoxifen, vary by the highly polymorphic cytochrome P450 (CYP) 2D6 enzyme. We prospectively investigated tamoxifen efficacy by serum endoxifen levels and the tamoxifen activity score (TAS).
1 sty 2018 · Tamoxifen (ICI 46 474), trans-1- (4-β-dimethylaminoethoxyphenyl)-1,2-diphenylbut-1-ene, is the most commonly used drug for the treatment of estrogen receptor positive breast cancer and has been saving lives worldwide for the past four decades.
Results. Women randomised to tamoxifen and reporting hot flushes at the 6-month follow-up visit had a non-statistically significant increase in breast cancer compared to those without hot flushes (HR = 1.26 (0.98–1.62), P = 0.08).
28 lis 2017 · Although tamoxifen has been on the market for several decades and is the most used drug against breast cancer, its use may still be improved. The present study shows that tamoxifen metabolism may predict breast cancer outcome by measuring serum concentrations of active tamoxifen metabolites.
