DOS 3.0 was the last major redesign of the DOS kernel. The basic structure of DOS remained largely unchanged since then, and most of the DOS 3.0 internal routines still exist in DOS 4/5/6 with only relatively small modifications.

MS-DOS 3.00. MS-DOS 3.0 debuted with the IBM PC/AT (as PC-DOS 3.0) and adds support for 5.25-inch, 1.2 MB floppy disks, support for the AT's internal clock and officially supports partition sizes up to 32MB, with sizes this large only being available from certain OEMs in prior versions of DOS.

DOS 3.0 adds support for FAT16 partitions up to 32MB, 1.2MB floppy drives, and the IBM AT internal clock. MS-DOS 3.0x is extremely rare. After the release of IBM PC-DOS 3.0, Microsoft dragged their feet providing MS-DOS 3.x to other OEMs, possibly to appease IBM.
