Chlorophyll plays an important role in the absorption of light energy of the plant. Chlorophyll can be extracted with a range of solvents. After extraction of the chlorophyll pigments with 80% acetone or 100% methanol, they can be measured in the spectrophotometer.

- Determination of Chlorophylls and Carotenoids by HPLC
This is the method that most plant scientists apply for the quantitative determination of chlorophylls a and b in plant leaf extracts obtained with organic solvents such as acetone or methanol. This method also allows to simultaneously determine the level of chlorophylls a and b and that of total carotenoids in the same leaf extract solution.
21 gru 2021 · In this paper we propose a fast, accurate and (almost) error-proof method to measure Chl a and b concentrations, as well as total Car content, in a solution containing pigment extracted from tissues, membranes or purified complexes. The method is based on the fit of the absorption spectrum of the acetone extract using the spectra of purified ...
A universal set of equations for determining chlorophyll (Chl) a, accessory Chl b, c, and d, and total Chl have been developed for 90 % acetone, 100 % methanol, and ethanol solvents suitable for estimating Chl in extracts from natural assemblages of algae.
4 gru 2020 · There are some methods to estimate the content of chlorophyll, such as based on the absorption of light by aqueous acetone extracts, based on the absorbance or reflectance of certain wavelengths of light by intact leaves using handheld chlorophyll meters.

As a rule, the chlorophylls must be extracted from the plant material before they can be estimated precisely (38, 193, 194). With clear plant extracts, the green pigments may be estimated directly by fluorimetry, colorimetry, or spectrophotometry (63, 65, 122, 123, 124, 125, 195).
