Phenylethyl alcohol agar (PEA) is a selective medium used to cultivate Gram-positive organisms, particularly cocci, from a sample containing a mixture of pathogens. The active ingredient, phenylethyl alcohol, inhibits or markedly reduces the growth of Gram-negative organisms by interfering with DNA synthesis.
Incubation. Incubate plates for 24 to 48 hours at 35oC ± 2oC in an appropriate atmosphere. In some cases, a longer incubation, up to 1 week, may be needed. PEA blood agar plates can be incubated under aerobic, anaerobic, and 5% CO2 atmosphere based on the type of microorganisms being studied.

SUMMARY AND EXPLANATION. Lilley and Brewer developed Phenylethyl Alcohol Agar for isolation of gram-positive cocci, such as staphylococci and streptococci, from specimens containing mixed flora.1 This medium permits the growth of gram-positive organisms while inhibiting most gram-negative organisms, especially the swarming of Proteus species.
Phenylethyl Alcohol Agar is formulated as per Lilley and Brewer (1) for the selective isolation of gram-positive bacteria. This medium can be supplemented with 5 % sheep blood.
