Unlike other calculators, the NEB Tm Calculator takes into consideration buffer components that affect melting temperatures and empirical observations when calculating the optimal annealing temperature.
Annealing temperatures can be optimized by doing a temperature gradient PCR starting 5°C below the calculated T m. The NEB Tm Calculator is recommended for calculation of an appropriate annealing temperature.
