These cells making up the mesophyll layer contain chloroplasts, thus making it the primary site of photosynthesis in green plants.

The spongy mesophyll cells contain air spaces to allow the movement of gases (i.e. carbon dioxide and oxygen) throughout the leaf. The guard cell helps to regulate the movement of gases into and out of the leaf. They also help to control the loss of water vapour (transpiration). The stomata are found in the lower epidermis.

Mesophyll cells (both palisade and spongy) are packed with chloroplasts, and this is where photosynthesis actually occurs. Epidermis also lines the lower area of the leaf (as does the cuticle). The leaf also has tiny holes within the epidermis called stomata.
