Given my too-close familiarity with my first VN-in-progress, and the variable nature of reading speed, I have no idea how to honestly estimate the length of my visual novel when someone asks how long it is. Is there a good rule of thumb to use, like "an hour per 1000 lines of dialogue" or something of that nature?

I would recommend doing the images as large as you can during production and then downscaling it to 4K (or larger if you want to allow zooming) . While most people do not have 4K monitors currently, it allows you to future proof your game better.
