Paper chromatography separates pigments in leaf cells on the basis of three criteria: Solubility; Molecule size; Polarity; Solubility is a measure of how well a pigment molecule dissolves in the solvent. In this project, the solvent is alcohol. Crushing the leaves breaks open cells so pigments interact with alcohol.
Try this class practical to use paper chromatography to separate and investigate the pigments in a leaf. Includes kit list and safety instructions.
1. Cut chromatography paper into a strip that will fit into the graduated cylinder. Cut one end of the chromatography paper into a point or tip. Make a light pencil line about 1.5 cm from the end of the paper you just cut. 2. Use the coin to extract plant pigments from the leaf.
