The Ruler tool calculates the distance between any two points in the workspace. Page 63 Columns help you position images or elements precisely. The New, Image Size, and Canvas Size commands let you specify image width in terms of columns.
You can use the Ruler tool (type "I", or "Shift-I" to toggle through until you've reached the Ruler tool) to plot a line between two points in the image. You should see measurements in pixels next to the tool details (below the menu items).
